If you are sure you have no blocked jets in the carb, it is time to look at the reeds and crank seals.

Ok sounds stupid on my part to even think it but I've seen once only someone drop the needle down the spout then put the slide in on top. I'm sure you wouldn't do 'that'. So forget that & try removing the needle & fit the slide without it in which case it should go super rich--just to prove a point.

& as Tinkicker says, check reeds. Any broken reeds like a big pieces missing. Think you replaced jets with similar but not used a long main jet & it's sitting against the bowl base.

You mentioned air screw & fuel screw but one just lifts the slide same as opening the throttle & other on an angle 'is' an air jet adjustment screw on the pilot jet. But no matter so shut up 'me' lol. Let's get it revving.

The fuel screw (engine side) enters in a angle to the pilot jet "hole" inside the carb, 1&half turns +- it gets the best idle. The air screw needs to fe fairly tight or won't idle like 1&half turns also. So maybe just maybe do you thing i put the float to low so its getting little fuel and as soon as the needle goes up its to much air for little fuel and it dies?

Pictures and a video of it running would help to find you a solution. You have to use a third party Imgur, google photos or YouTube etc

Not piston in backwards!! with skirt holes towards exhaust. Have seen that before & if you got it running it'd be all up the creek & hollow gaspy needing choke most likely.
