how far can a 250LB rider go on a dt360 up a 10% average grade?

I think you will have no issue. I doubt you'll even hit reserve.

Mine gets 58 mpg overall on my daily commute, and we do have hills.

A buddy of mine has a '72 360. We do a ride out of Livermore, CA on occasion that is about a 38 mile one-way pavement ride, that goes back in to the hills. His bike is good for about 45-50 miles on this ride, so we always bring a tank. For your Santiago peak ride I'd take a small amount of spare fuel with you in a backpack the first time, just for your "test" so to speak. Knowing that ride, my guess is you won't need it.

MSR fuel bottles work good for carrying spare some fuel. They make holders for them that attach with velcro straps to the frame.

I'm still guessing about half a tank for the round trip if ridden at a leisurely pace. If it's a full throttle race to the top, that would be different.

Better safe than sorry.

I have hiked Santiago via Holy Jim, but its been closed since the maniac lit it on fire a few years ago.
"Racks" is a respectable hobby :-) i will see what i can find

So i moved here from Chicago 6 years ago and dont have a Motorcycle endorsement on my DL (been flying under radar), but did get my new project titled, plated and registered in 48 hours with a service for $250 if you could believe that. Does anyone have a tip on the quickest way through the required "motorcycle safety class" so i can take the test and get this?

The safety course here in Texas is very rudimentary, cross your tees and dot your I , most of us could teach it , a few hours of boredom and your out
